In Cervical Cancer Screening, koilocytosis is the most recognizable criterion for the presence of HPV.

CytoProcessor® shows the presence of these koilocytes, on a Liquid Based Cytology slide, at a first glance, thereby assisting the screener in making an accurate and reliable diagnosis for LSIL.

Did you ever wonder what the mystery is behind this cytopathological phenomenon that’s transforming normal squamous cells into these characteristic koilocytes?

Well, koilocytes can develop when the HPV virus is transmitted through sexual contact in cases where the immature basal cell layer is no longer protected due to damage to the protective layer of mature cells above it.

The sexually transmitted HPV DNA virus, which transcribes into DNA strands after infecting a basal cell, leading to the formation of viral proteins E6 and E7, which are responsible for the oncogenic effects of the virus. It multiplies in the maturing squamous cells as the virus incorporates its DNA into the genome (= all 23 pairs of chromosomes in the cell). A fusion of different proteins is ultimately responsible for the formation of the large cavities that are so characteristic of infected HPV cells.

Microscopically characteristics of koilocytes
Mature squamous cells with:

  • Large clear irregular cavity or halo with sharp border

  • Enlarged, slightly atypical nucleus and almost never in the centre of the halo

  • Bi- and multi-nucleation is common

IMPORTANT: Digital Cytology starts with an optimal sample collection (NO LUBRICANTS) followed by excellent standardized preparation into Liquid Based Cytology slides, standardized staining of the LBC slides and cover-slipping to make an accurate and reliable diagnosis.

This case example shows CytoProcessor® can be used with Hologic ThinPrep or ILSA Diagnostics preparation, iDC20  and CellExpert or  BD Surepath (only RUO) and a Hamamatsu S360 or 3DHistech P250, P480 and P1000, Leica GT450, EVIDENT and ARGOS 6X scanner.
